Posaconazole Impurity 53(CAS#149809-43-8)
Introduction
(5R-cis)-toluene-4-sulfonic acid 5-(2,4-difluorophenyl)-5-(1H-1,2,4-triazole-1-yl)methyltetrahydrofuran-3-ylmethyl ester, also known as TMSOTf, is a commonly used reagent in the field of organic synthesis. The following is an introduction to its properties, uses, manufacturing methods and safety information:Quality:- Appearance: Colorless liquid- Solubility: Soluble in most organic solvents such as ethanol, ether and dimethylformamideUse:TMSOTf has a variety of applications in organic synthesis, including but not limited to:1. As a strong acid catalyst: TMSOTf can be used as an acid catalyst in a variety of reactions to promote the etherification, esterification, phosphorylation and other reactions of ketones.2. As a dehydrating agent: TMSOTf can be used as a dehydrating agent under non-aqueous conditions, and is often used in the dehydration reaction of ethers, alcohols and other compounds.3. As a sulfonic acid protecting group for aromatic compounds: TMSOTf can react with aromatic amines to form sulfonate-containing products, thereby protecting amine groups.Method:The preparation of TMSOTf is usually carried out by the following steps:1. (5R-cis)-toluene-4-sulfonic acid is reacted with dimethylformamide to obtain (5R-cis)-toluene-4-sulfonic acid dimethylformamide ester.2. Then, (5R-cis)-dimethylformamide toluene-4-sulfonate is reacted with trimethylsilyl trifluoromethanesulfonate (TMSCl) to produce (5R-cis)-dimethylformamide toluene-4-sulfonate trimethylsilane.3. The (5R-cis)-toluene-4-sulfonate dimethylformamide ester trimethylsilyl ester was deprotected by cesium fluoride (CsF) or trifluoromethanesulfonamide (TfNH2) to remove the final product TMSOTf.Safety Information:1. TMSOTf is irritating and corrosive to the skin, eyes, and respiratory tract and should be avoided.2. Wear appropriate protective gloves, safety glasses and protective clothing when using.3. Operate in a well-ventilated area and avoid inhaling steam.4. Keep the container tightly sealed and place it in a cool, dry place, away from fire.
